Our research aims to tackle the challenges in the functional characterization and biomarker development of extracellular vesicles (EVs). We focus on improving pre-analytical procedures by stabilizing EVs and isolating specific EVs from other particles. Reliable characterization of EVs is essential, so we develop and refine measurement techniques, procedures, and software to accurately analyze EVs. We study their biochemical properties, like antigen expression or miRNA content, functional properties, such as procoagulant activity, and physical properties, including size and concentration.
Our work requires pure EV samples, creating a strong synergy between pre-analytics and EV characterization research lines. To ensure reproducible data, we aim to standardize all assay aspects. Measurement techniques often use arbitrary units, making standardization challenging. To address this, we collaborate with European metrology institutes to develop reference materials and procedures that express EV measurement results in standard units with known uncertainties. This standardization allows us to validate pre-analytical procedures and measurement techniques and reproduce results.
Situated in the largest medical center in the Netherlands, we apply these standardized methods in clinical research to search for EV-based biomarkers.
