Advanced skin tissue culture systems are an essential technology, driving basic and disease-relevant investigations in cutaneous biology. Among the most powerful tools are 3D reconstituted human skin (RHS; also called organotypic skin or raft cultures), derived from primary cutaneous cells. However, these culture methods require considerable expertise and access to fresh tissue, or are associated with significant cost from commercial sources. The Skin Tissue Engineering and Morphology (STEM) Core of the Skin Biology and Disease Resource-based Center (SBDRC), funded by the National Institutes of Health, will provides reliable, state-of-the-art technology, tissue, cells, materials, and expertise for human and mouse skin culture models. We are a source of normal and diseased human skin tissue for the derivation of keratinocytes, fibroblasts, and melanocytes for application in 2D and 3D human skin cultures. We also isolate mouse keratinocytes and fibroblasts for investigators with transgenic or knockout mouse models that have a skin phenotype for use in generating 2D and 3D cultures. The STEM Core serves the SBDRC as a continuous pipeline to provide skin cultures and to assess their morphology and function by offering a variety of services and instruments for evaluation. The STEM Core also maintains a repository of skin cells and ex vivo tissue to assist investigators in translating their basic research findings. We provide in-depth training to facilitate proficiency in all skin culture methodology and imaging technology. We expect to be a vital resource for biomedical researchers by providing critical services and training that will galvanize investigators to address important questions about normal and disease states in investigative dermatology.
