Project: "Differential modulation of the p53 network in cancer therapy"I have engineered a transgenic mouse model, carrying an extra copy of the cell cycle regulator gene Cdkn1a (p21), to investigate p53-dependent mechanisms of tumor suppression. Mice with three p21 alleles (Cdkn1aSUPER) show an increased cell-cycle arrest capacity with a concomitant protection from DNA damage-induced apoptosis. Of note, The Cdkn1aSUPER mice display delayed epithelial regeneration and are robustly cancer resistant finally indicating p21 and p21-regulating pathways as potential drug targets (Torgovnick et al., Cell Rep., 2018). The Cdkn1aSUPER mice are now available from the Jackson Laboratory mice repository.
