Ph.D.Chatrapati Sahuji Maharaj Kanpur University, Kanpur Jan 2000 - Feb 2008Up, IndiaDuring my Ph.D. I used various DNA Fingerprinting techniques to study diversity among various species of aromatic grasses. I had been trained in various molecular biology techniques like from DNA/RNA isolation, qPCR, cloning, sequencing, hybridization, primer designing, molecular marker development etc. In brief I used to isolate DNA from different varieties and species of aromatic plants (Cymbopogon and Vetiveria) for doing PCR with arbitrary primers (designed inhouse) in order to study the degree of diversity among them through phylogenetic tree, component and Clustal W analysis. Further analysis was followed by cloning and sequencing of the selected co-migrating fragments through Sanger’s chain-termination method using fluorescent tagged dideoxynuleotides (ddNTP).
