R&D Scientist (Advanced)Agilent Technologies Feb 2019 - PresentSanta Clara, Ca, Us Invented a patent-pending 96-well plate-based one-pot release and labeling technology for the rapid high-throughput preparation of O-glycans obtained from glycoproteins and analysis by LC-MS. This technology is the fastest, most sensitive, and high-throughput workflow for this analysis to date. Designed, invented, and synthesized a novel fluorescent dye as an orthogonal label for O-glycans. This new dye is to be implemented for LC-FLD-based applications in combination with optional mass spectrometry detection which fulfills the current market need for O-glycan analysis. Evaluated numerous release and labeling techniques to establish feasibility of a commercial O-glycan analysis kit. These methods include classical Carlson degradation, hydrazinolysis, and one-pot based release/labeling for analysis by LC-FLD, LC-FLD-MS, and/or LC-MS platforms. Assembled, managed, and directed a project team of one scientist and two research associates for the development of a commercial O-glycan analysis kit. Managed and directed bi-weekly team project meetings to ensure productivity towards the development of a commercial O-glycan analysis kit and evaluate the research performance and growth of each attendee. Attendees include two scientists and two research associates. Managed and administered training to two scientists and three research associates in the utilization of all Agilent instrumentation and associated software. Supervised a scientist and two research associates on additional non-O-glycan projects which include intact protein analysis, BRAVO automation, sialic acid quantitation kit product requirements, instantQ purification, customer and marketing support, and general duties. Co-developed workflow procedures for Agilent’s AdvanceBio Gly-X APTS Express kit which resulted in its commercialization in 2019. Provided support to Agilent/ProZyme sales and marketing teams regarding customer-related questions regarding the ProZyme and Agilent portfolio. -
R&D ScientistProzyme, Inc. - A Part Of Agilent Jun 2018 - Feb 2019 Played a key role in the acquisition of data needed for the commercialization of two ProZyme products: APTS Express and sialic acid quantitation kit. Collaborated with Richman Chemicals for the development and evaluation of dyes for improved glycan resolution in the ProZyme Gly-Q CE system. Independently and successfully site-prepped the Agilent Hayward site for a total of five Agilent instrumentation after its acquisition by Agilent. This involved preparation and installation of two Agilent LC/FLD systems in addition to an Agilent LC/MSD XT, Agilent LC/TOF MS, and Agilent LC/QTOF MS system. Site preparations include pump exhaust plumbing, nitrogen gas generators and associated plumbing, and electrical outlet installations. Transferred workflows and optimized the Agilent instrumentation to work in conjunction with the ProZyme product portfolio. Managed and administered LC and LC-MS training to research assistants and scientists. Designed and directed research projects to a team consisting of three ProZyme research assistants. Successfully optimized and developed protocols for analysis of N- and O-glycans produced/labeled using both pre-existing and future ProZyme products on LC/FLD, LC-MS, and Gly-Q CE systems. Developed workflows and protocols for the LC and Gly-Q CE analysis of glycans labeled using exploratory dyes containing “instant” chemistry.
