About Adam Spaulding, Phd
Microbes are at the very foundation of virtually every process in life - from primary production, biogeochemical processes, to human health. Notably, bacteria are key players. I am very passionate about studying bacteria to better understand these organisms so that we many take advantage of them to enhance human lives and the environment. I became interested in microbiology, specifically bacteriology, as an undergraduate. I found the complexities and elegant systems within the bacterial world to be fascinating. Learning their impact on the Earth and its ecosystems drove me to further study them.I have spent the better part of my research career trying to understand how bacterial pathogens cause disease and develop vaccines and therapies against these infections. In doing do, I have come to have a better appreciation for just how much bacteria can do to actually help people as opposed to harm them.I am passionate about working with a diverse team of people of all backgrounds to exploit bacterial systems in order to develop new therapeutics, new vaccines, new agricultural practices and new ways of producing energy to benefit all of humanity.

Studied how the bacterial pathogen, Clostridium difficile, regulates certain factors important for the colonization of the human gastrointestinal tract and subsequent infection. Clostridium difficile is the most common cause of nosocomial infections and is the cause of approximately 29,000 deaths in the United States each year, and the economical burden is estimated between $3 billion USD and $5 billion USD (Lessa et al. 2015. NEJM -http://www.nejm.org/doi/full/10.1056/NEJMoa1408913). I was particularly interested in how a bacterial second messenger called cyclic diguanylate (c-di-GMP) regulates gene expression through structured RNA regulatory domains called riboswitches. Importantly, C. difficile contains a number of these riboswitches upstream of a variety of genes, but for our work, there is a riboswtich upstream of the flgB gene, the first gene of the early stage flagellar operon in C. difficile. Rita's group showed that c-di-GMP negatively regulates this gene. We hypothesize that this negative regulation occurs directly through the riboswitch upstream of flgB. In order to test this hypothesis, I developed a number of different reporter plasmids, and I generated mutations in the different active sites of the riboswitch to alter activity. I worked on developing a number of other genetic tools for C. difficile, including an allelic exchange plasmid, because the tools for genetic and molecular modification of this organism are lacking.
