These seeds were also at the different growth stages and exposed to different conditions--room temperature, cold temperature, salt conditions, and a combination of salt and temperature. This was done by using a drill pestle, and using various rinses (2M NaCl, SDS, sterile water, acetone) to be able to extract pure starch. The starch samples were then examined under the microscope to look at the alpha and beta granules to see how starch is broken down at the different growth stages. Iodine staining was also used, and merbromin staining was used on select dried starch samples.
