Obesity represents a major independent risk factor for myocardial infarction and heart failure. Yet, the effect of obesity on the outcome of acute coronary syndrome is controversial with clinical studies suggesting a paradoxical favorable effect. Previous results from our group indicate that high fat diet induced obesity protects against acute myocardial ischemia/reperfusion injury. In this project, we aim to determine how obesity modifies myocardial glucose uptake and glucose metabolic pathways and the contribution of these changes to the cardioprotection observed in obese mice. We will work with a mouse model of high fat diet-induced obesity and subjected to transient coronary occlusion. Infarct size and functional data will be measured at the end of the experiment. Metabolomic analysis will be performed by 1H-NMR measurements and variations in metabolic pathways by the administration of 13C-labeled substrates combined with 13C-NMR. Proteins involved in glucose transport will be quantified by western-blot and PCR.

Establish the nitrogenous needs and preferences of different yeasts, not Saccharomyces and Saccharomyces, growing curves with a plate reader in order to determine the limiting nitrogen concentration and the preferred sources for each strain. Once these conditions are established, individual fermentations with synthetic beans will be made to see the nitrogen consumption profile of each one of the species during fermentation, as well as their growth and viability. The fermentations will be monitored by density / consumption of sugars and the viability of yeasts throughout the process. The consumption of amino acids and ammonium will be analysed by means of HPLC.
