Investigating new uptake routes of iron in human cells

Project title: Antimalarial drug discovery using a target-based platform.The fully automated antiparasitic drug-screening method was used selecting compounds specifically targeting parasite enzymes. Promising ones with antiparasitic activity have been tested with the use of yeast-based platform (bioscreens). The compound toxicity has been tested on eukaryotic cell lines. The inhibiting effect of compounds is measured with a plate reader and the kinetics is analyzed/optimized. Among the experimental tasks performed during the project were: enzyme inhibition assay, cytotoxicity assay, mammalian cell culture.

The C terminus of mammalian TERB1 protein is composed of the TRFB domain and a MYB-like (MYB) domain. The C-terminal MYB domain of TERB1 is thought to be a distinct functional unit that facilitates telomere rigidity through the recruitment of cohesin complexes to the meiotic telomeres. The project involves experimental laboratory investigations on the KO mouse model abolishing MYB-like (MYB) domain expression. Among the experimental tasks performed during the project were: handling mice, tissue processing for histopathology, meiotic chromosome spreads, preparation and immunostaining, fluorescence microscopy, Q-FISH,bacterial clonning, genotyping, protein expression and purification, western blot and co-immunoprecipitation.

Project title: Investigation of redox-sensitive thiol in Schizosaccharomyces pombe MAPKK Wis1.The project focused on the oxidation of Schizosaccharomyces pombe MAPKK Wis1 C458 which inhibits kinase activity and this inhibition is required for an appropriate pathway response to peroxide. The results obtained during the project suggest the presence of a thiol-redox inhibitory mechanism. Among the experimental tasks performed during the project were: bacterial clonning, yeast transformation, PCR, spot test, biochemical assay for monitoring protein thiol redox states, protein expression and purification, western blot.
