LIfE is an integrated life-detection package that combines automated microfluidic sample processing with brightfield and epifluorescence microscopy to identify and characterize morphological and textural indicators of life and to identify, resolve, and characterize microscale structural features indicative of cells and cell fragments. • Perform research to assist with the characterization & calibration (C&C) and verification & validation (V&V) of the instrument’s main subsystems: sample transfer, fluidics, optics, and electronics and software. • Work will focus on optics (bright-field and fluorescence microscopy) C&C and V&V using a variety of bacterial and archaeal type strains with known characteristics, as well as environmental samples possessing properties expected in Ocean World ice materials (e.g., polar ice, hypersaline brines, and oligotrophic marine and groundwater systems). • Use results to define and optimize instrument operation protocols, excitation and emission wavelengths, and fluorescence stain selection for future flight opportunities.
