My research demonstrated how antibody immunogenicity can be reduced by humanising CDRs via germline substitutions without loss of binding (Augmented Binary Substitution; ABS). We published the results in Nature Scientific reports and the technique was integrated into pipeline therapeutic development. At Evotec I carried out research as a Protein biochemist specializing in X-ray crystallography, protein engineering, and three-dimensional protein structure calculations. With a proven track record of collaborating on multidisciplinary projects, I translated intricate molecular insights into practical applications for therapeutic development.I leveraged cutting-edge experimental and bioinformatics techniques to decode the complexities of protein structures in a computer-aided drug design approach. About the team: Evotec’s Structural Biology department specializes in a wide range of structural biology services, including the study of soluble and membrane proteins, protein-protein and protein-DNA/RNA complexes, and RNA molecules. They utilize advanced techniques such as synchrotrons, cryo-EM, and X-ray crystallography to support drug discovery and development. The crystallography team is fully integrated into the Evotec structure-based drug discovery workflows for a better understanding of the molecular interactions, fast structure turnaround and reduction of design cycles.

The focus of my postdoctoral research was the structural study of membrane proteins involved in drug transport and nutrient uptake. Our research in collaboration with Professor Simon Newstead (Oxford University) showed how membrane transporters involved in nutrient uptake altered their three-dimensional protein structure to accommodate different sized peptides. This is important to enable the design of drugs that exploit this to cross cell membranes in disease treatment pathways. We published our results in the high-impact journal Proceedings of the National Academy of Sciences (PNAS) and contributed three co-crystal structures to the global Protein Data Bank (www.pdb.org).
