About Alexander Brean
🎓 PhD in Biochemical Engineering | 🔬 Medical Science LiaisonAn enthusiastic scientist with 10 years of laboratory experience across a wide range of diciplines. I have just started as a Medical Science Liaison. In my lab-based roles, I focusd on solving problems encountered in manufacturing. This can be anything, from antibody activity, antigen excess, curve activity, precipitation, raw material issues, non conformances, etc. During my PhD, I was involved in hydrophobic interaction chromatography; using temperature to change binding/elution, rather than mobile phase composition. This was successfully employed for both BSA, and monoclonal antibodies.

Mres Project Student
Project 1: Under the supervision of Professor Grant Stewart's Laboratory, I explored the role of DONSON in the DNA damage response and cell cycle regulation. Utilising techniques such as immunoprecipitation, Western blot analysis, fluorescence activated cell sorting, fluorescent microscopy, and human cell culture, I identified proteins interacting with DONSON. My contributions to this project have led to a publication in Nature Genetics.Project 2: In Dr. Aga Gambus's Laboratory, I investigated MCM7 polyubiquitylation in DNA replication fork termination using Xenopus Laevis as a model organism. This project enabled me to learn vital techniques, including chromatin isolation, working with X. laevis egg extract, bacterial cloning, PCR site-directed mutagenesis, and protein purification. Through this research, I gained a deeper understanding of the effects of blocking MCM7 polyubiquitylation on DNA replication fork termination.
Placement Student
At the University of Plymouth I characterised the unique properties of mouse alveolar macrophages when exposed to IL-4 and IL-13. My responsibilities included culturing alveolar macrophages, preparing whole cell lysates, and performing 2D SDS-PAGE separation, which facilitated the subsequent mass spectrometry analysis. This experience not only honed my skills in advanced laboratory techniques but also provided valuable insights into the intricacies of cellular research and collaboration in a scientific setting.
