I am a PhD candidate in the University of Cincinnati's Neuroscience Graduate Program. I currently work in the Davidson Lab in the Dept. of Anesthesiology. My dissertation research focuses on the contributions of skin-nerve interaction to pain and itch, with a focus on atopic dermatitis. In order to answer my research questions, I use both in vitro and in vivo models of inflammatory skin disease and have developed protocols for methods such as calcium imaging, patch-clamp electrophysiology, ELISA, qRT-PCR, immunostaining, and H&E staining. In addition to my dissertation work, I am involved in past and present projects in the lab related to the peripheral nervous system, including investigation into cannabinoid receptor CB1 expression and function in human DRG sensory neurons, transcriptomic and functional comparison of human iPSC-derived sensory neurons with native human DRG sensory neurons, and testing the efficacy of an experimental naloxone prodrug developed by the Merino lab in Chemistry. I also play a very active role in a number of collaborations. Central to these is our collaboration with the Supp and Schutte labs in the departments of Surgery and Biomedical Engineering, respectively, to develop an all-human-cell 3-dimensional in vitro model of innervated human skin. When successful, this engineered skin will have immense utility, in both clinical and research applications. I also played an important role in a collaboration with the Hershey lab at Cincinnati Children's Hospital Medical Center in which I designed and conducted experiments to characterize sensory neuron sensitivity in their mouse model of atopic dermatitis. Finally, I have been responsible for conducting experiments on a collaboration with the Tao lab at Rutgers which are designed to assess the effects of manipulating a long noncoding RNA on human DRG sensory neurons using in vitro models of chemotherapeutic and diabetic neuropathy.My research experience and education has prepared me for my long-term career interest in medical laboratory science. I look forward to a career in which I can apply my laboratory, problem solving skills, and project management experience to inform proper patient care.

Responsibilities varied, depending on the lab, but I was always responsible for learning new techniques and applying them to run experiments related to a given project, as well as performing data analysis and interpretation and routine laboratory maintenance. During my first rotation, I was tasked with and was successful in implementing a western blot protocol for general lab use.Techniques: western blot, Nanodrop nucleic acid quantitation, qRT-PCR, RNA purification, plasmid purification, mouse behavioral test scoring, H&E stain quantification, immunohistochemistry, immunocytochemistry, widefield fluorescence microscopy, cryostat sectioning, dorsal root ganglia neuron recovery and primary culture, cAMP imaging, human embryonic stem cell culture, super-resolution microscopy, FRET, routine lab maintenance, buffer and reagent preparation, statistical analysis.

While in the Gross lab at CCHMC, I worked on a project studying the effects of microRNA antagonists on the expression of the A-type potassium channel subunit, Kv4.2, as well as different facets of electroencephalographic (EEG) readouts, seizure susceptibility, neuronal degeneration in the hippocampus, and dentate gyrus (DG) neuronal migration in different mouse models of temporal lobe epilepsy.Additionally, I worked on a project aimed at elucidating the effects of altered Kv4.2 expression on the expression of its associated subunits, as well as its effects on EEG characteristics.Skills which I have used and learned during my time in the lab include PCR, tissue lysis, DNA and protein gel making, western blotting, buffer making, mouse genotyping, basic mouse handling, cryostat section of mouse brain tissue, histology on mouse brain tissue (Fluoro-Jade B, Nissl staining, fluorescence immunohistochemistry), widefield brightfield and fluorescence microscopy, confocal fluorescence microscopy, quantification of stained mouse brain tissue using ImageJ or NIS-Elements software, EEG analysis using NeuroScore software, researching and optimizing procedures, and routine laboratory maintenance.

I worked on DNA sequencing of tissue samples of northern mockingbirds (Mimus polyglottos) from different parts of the North American continent in order to determine if the species had diverged into different subspecies. The overall DNA sequencing procedure involved making DNA gels, PCR, gel electrophoresis, and the use of a DNA sequencer. In addition to bench work, I was expected to be able to analyze and interpret data.
