Team A.TurtoïI am using several methods of molecular biology and yeast display to develop new bispecific antibodies with "sweeping" mechanism in order to enhance treatments of digestive cancers.Je travaille sur le développement d'anticorps bispécifiques dits à activité de sweeping destinés à améliorer le traitement des cancers digestifs.

I was in charge of potential allosteric drugs screening with in vitro functional assays using fluorescent and radioactive tagging. To ensure that testing and analytical conditions were equivalent, we had to follow strict quality processes in particular with 384 wells based experiments. As some of our assays were based on living cells, I was routinely taking care of various cell lines. Addex promotes communication around all projects which permitted me to have a good overview of all the pharmaceutical development processes.

I was in charge of flow cytometric products with unmet specifications to identify and solve these problems which usually require ensuring departments’ cooperation. In order to identify the root cause of the problems, I have imagined my own experiments to test hypotheses and driven investigation through products’ lives. Then, I had the responsibility to determine if we can save the product and if we should save it still strictly following quality requirements. To finish, I had to facilitate interpersonal communication by making clear summaries of my work to present it to departments which made the final decision. J'ai travaillé 8 mois dans le service Ingénierie d'Immunotech Beckman Coulter.Mon activité était centrée sur des produits de cytométrie en flux dans le cadre de non-conformité. Mon rôle était donc de vérifier la qualité des produits et d'assurer le respect des spécifications.Cette expérience a été très enrichissante puisque j'ai pu découvrir le fonctionnement d'une large gamme de produits de cytométrie en flux, interagir avec l'ensemble des services pour résoudre les problèmes, organiser des réunions et rédiger des rapports détaillés ainsi que des documents de suivi.

I initiated a project whose goals were to access anti-TNF drugs’ mechanisms and donor heterogeneity thanks to a flow cytometer panel. I began with a consequential bibliographic work to set targets of interest, and then I made and adjusted lots of cytometric experiments. This work permits me to learn a lot about autoimmune disease and inflammatory mechanisms and treatments. We used blood samples with different preparations to mimic physiological or pathological conditions. I analyzed results of my multi-parametric panel of flow cytometry, discussed the next experiments with my internship mentor, setting my experiment schedule and made a completed detailed report of my whole work. Au cours de mon stage de fin d'études, j'ai intégré l'équipe de R&D Life Sciences d'Immunotech (Beckman Coulter). La mission de mon stage consistait à travailler sur un test de détection en cytométrie en flux. Ce stage a été très instructif puisque j'ai pu me familiariser avec la cytométrie en flux, qui est un outil complexe mais puissant. J'ai également beaucoup développé mon autonomie grâce à la confiance que m'a accordée M.Busnel.

I have integrated the Schmolke group whose work is focused on influenza viruses. I made lots of cell culture in a L2 lab to be able to work with viral material and test protein-protein binding. This experience was very complete since I made DNA extraction and amplification by PCR, plasmid design, transfections, sequencing, protein analyses by Western Blot and immunofluorescence. It was also a real pleasure to work in this international environment which permits me to discover other cultures. I really have appreciated to carry my experiments and analyses independently and then have the opportunity to discuss my results with my mentor and the research team to discover another point of view and benefit from their advices.Au cours de ce stage, j'ai eu la chance de rejoindre le groupe de recherche du professeur Schmolke ce qui m'a permis d'apprendre beaucoup de choses sur les virus. Durant ces 10 semaines, le docteur Anchisi m'a permis de participer à de nombreuses expériences et ainsi j'ai pu développer mes compétences de cultures cellulaires mais aussi acquérir une bonne expérience des manipulations courantes dans la recherche virale et découvrir les règles de sécurité encadrant la manipulations de virus dans un laboratoire de recherche. J'ai également pu me faire une idée de ce qu'est l'organisation d'un groupe de recherche et de ce que signifie travailler dans le monde de la recherche au quotidien.
