About 70% of plants, including many crops such as wheat and rice, enter a symbiotic mycorrhizal association with a fungus. During this association, plant roots gives to the fungus sugars and get in return water and mineral nutrients. In order to initiate this symbiosis, both organisms enter into communication and need to recognize their partner in order to avoid any defense response.During my premaster project, I was investigating rice proteins that might have the ability to detect fungal cell walls. To do so, the proteins of interest were down-regulated by RNA interference (RNAi) and my first goal was to check the effectiveness of this down-regulation by diverse molecular techniques. The second aim was to observe a potential effect on root mycorrhiza capacity of these rice mutant plants by mycorrhizae staining and quantification.This project was done under the supervision of Dr. Uta Paszkowski, Department of Plant MolecularBiology (DBMV), Lausanne, Switzerland.Skills:- Southern blot- RNAi- RT-PCR- RNA extraction- Rice cultivation- Mycorrhizae staining and quantification
