My thesis project has centered around using the fruit fly, Drosophila melanogaster, to genetically model nonsense-mediated mRNA decay (NMD). The NMD pathway functions to target and degrade certain cellular mRNAs, especially those mRNAs harboring detrimental mutational changes. NMD function is necessary for viability in humans and the fly, and it is thought that the overexpression of target mRNAs that occurs when NMD has been disabled leads to inviability. This implies that NMD provides a negative regulation on gene expression that is critical for cellular and/or organismal functions. My project has been to use various genetic, molecular, and bioinformatic techniques to identify direct mRNA substrates of NMD in fly, with a focus on identifying the specific mRNAs whose negative regulation is critical for viability.
