About Amanda J. Lessard, Ph.D.

Research Navigator with a history of working in academic and government research settings. Experience with project management to include research design, implementation, and completion by way of published work through both individual and team collaborations spanning multiple disciplines. Cell and molecular biologist by training in the fields of hematologic oncology and cardiovascular research with expertise in genomics, flow cytometry, primary human 3D scaffold-free spheroid cell cultures, protein biochemistry techniques and small animal studies. Current training and work have shifted focus to the regulations and policies involved with human subjects' research and project management of these clinically centered studies as well as community engaged research methodologies. Dr. Olgun Guvench’s at the University of New England School of Pharmacy learning biomolecular simulation and computer-aided drug design; specifically, investigating the interactions between hyaluronan and CD44. This resulted in a first author publication. My thesis dissertation research was performed within Dr. Sathyanarayana's laboratory at MMCRI understanding cytokine and epigenetic regulation of microRNA in acute myeloid leukemia. My thesis work encompassed various molecular biology techniques and technologies with immortalized and primary cell lines, multiple knockout mouse models, and human tissue samples.     The goal of my project was to define the roles of Podocalyxin (PODXL) in onset and progression of acute myeloid leukemia (AML). In recent parallel studies, several reports have associated increased PODXL expression with the advancement of several aggressive cancers including acute myeloid and lymphoid leukemias; as well as certain breast, liver and pancreatic tumors. Via employing cell line models, mouse models and clinical AML samples, investigations were performed to define PODXL’s role in development and progression of AML. In addition, my projects evolved into also addressing the role of miRNAs in AML due to several recent investigations having linked aberrant miRNA expression with AML.

    My role was as a member within the Genomics Unit of the Research Technologies Section in Hamilton, Montana run by Dr. Steve Porcella. I performed capillary/Sanger DNA sequencing for all intramural NIAID researchers at both the Rocky Mountain Laboratories campus and the main campus in Bethesda, Maryland. My collaborative work was recognized in multiple peer-reviewed journals noted below in the publication section but most notably was the New England Journal of Medicine publication with Dr. Helen Su and her colleagues. This research was recognized with all members receiving the NIH Directors Award for "identifying the genetic cause of a previously undefined primary immune deficiency disease".
