    StemCell Gene Editing TeamOur institute mission strives to understand the complexities and interplay between cell organization, behavior and identity in the context of dynamic environments - using gene-edited hiPSCs. The experiments I perform help us study and understand how cells interact and behave as they transition into another cell state and differentiate. Specific skills: -Passaging and maintenance of gene-edited hiPSCs-Differentiation of hiPSCs using small molecule and growth factor protocols into early mesoderm lineages-Gene expression quantification using ddPCR-iCRISPR interference-Live cell fluorescence imaging and image analysis using ImageJ/Fiji, Zen, Las X software-Flow cytometry using Cytoflex and data analysis using FlowJo -Immunochemistry and immunofluorescence staining techniques-Stocking and inventory lead

    Led two independent projects that further explored candidates for vaccine development, diagnostic testing, and genetic manipulation of T. pallidum TprL project: Tested human sera samples infected with Yaws and Syphilis for variability in TprL antigen reactivity using ELISA. The aim of the project was to examine if there were significant differences in reactivity to TprL, a putative OMP, in Yaws and Syphilis patients to evaluate whether TprL could be part of an antibody test for differential diagnostics. FlaB 1/3 project: Designed a plasmid vector that will be transformed into T. pallidum using a CaCl2-based transformation protocol. This project will involve the continuous in vitro propagation of T. pallidum, and the transformation process will allow knock-out of the flaB1 and flab3 genes, encoding the flagella core in T. pallidum, and replacement with both genes at once with a Kanamycin resistance cassette through homologous recombination. Successful knockout will allow us to further our understanding of motility as a T. pallidum virulence factor. I am proud to be part of the team that first discovered how to genetically manipulate this pathogen!
