I am currently located in Bar Harbor where I am wrapping up my PhD research at the Jackson Laboratory. I am one of three people in the world working on a rare form of congenital muscular dystrophy with megaconial myopathy (MDCMC), in which patients suffer from limb muscle degeneration, severe cognitive impairment and have giant mitochondria. My work involves a range of cell and molecular biology techniques, focus on different therapeutic strategies, including gene therapy (AAV injections) and dietary modifications, for the rescue of muscular dystrophy and a wide variety of behavioral testing for motor and cognitive impairments in mice. I have experience in Biotechnology, having interned at Biocon Ltd. and higher education, having taught courses in Genetics and Developemntal Biology at the undergrad level. I am looking to transition into industry post graduation this fall.

A novel disease mechanism identified in rostrocaudal muscular dystrophy (rmd)Rostrocaudal muscular dystrophy (rmd) is a new recessive mouse mutation that causes a rapidly progressive muscular dystrophy. We identified the rmd mutation in the choline kinase beta (Chkb) gene, resulting in a complete loss of CHKB protein and enzymatic activity. Choline kinase catalyzes the phosphorylation of choline to phosphocholine in the biosynthesis of the major membrane phospholipid phosphatidylcholine (PC). While mutant rmd mice show a dramatic decrease of CHK activity in all tissues, the dystrophy is only evident in skeletal muscle tissues in an unusual rostral-to-caudal gradient. Enlarged mitochondria are evident in rmd muscles, suggesting that dysregulation of energy metabolism may underlie the muscle disease. Our identification of the mouse Chkbrmd mutation directly led to the identification of similar human CHKB mutations in children diagnosed with a severe megaconial congenital muscular dystrophy (MDCMC). The rmd mutant mouse offers the first demonstration of a defect in a phospholipid biosynthetic enzyme causing muscular dystrophy and provides a unique resource for understanding the causes and potential treatment of this disorder.
