My Contributions:# Revealed the charged amino acids like Lysine, Glutamate and their capped (N/C termini) derivatives do show weak intrinsic luminescence when excited at the near UV and visible regions. These amino acids were reported to exhibit nominal extinction coefficient in this spectral region due to photoinduced charge transfer processes termed as Protein Charge Transfer Spectra.# Observed a weak intrinsic luminescence could arise from charge recombination (post excitation to charge transfer states) in monomeric proteins rich in charged amino acid residues.# Characterized the photophysical properties of such intrinsic luminescence from dense aqueous solutions of charge amino acids as well as monomeric proteins.# Explained that the luminescence from charge transfer states could affect the fluorescence lifetime decay of Tryptophan, probably explaining one of the reasons behind the multi-exponential decay of Trp in proteins.# Established the potentials applications of Protein Charge Transfer Spectra and luminescence arising from charge transfer states in tracking and detecting protein oligomerization/aggregation and protein unfolding.Technical Skills Developed:UV-Visible spectrophotometry, Fluorescence spectroscopy (stead-state and time-resolved), Time-resolved decay analysis (discrete and maximum entropy method), Protein expression and purification, Mass spectrometry (MALDI-TOF), Biochemical assays (Lowry test, ThT assay), Protein labelling with fluorescent probes like Dansyl, Cy3, Cy5, etc.
Visiting Research Scholar
I was a visiting research scholar for a collaborative work on the preparation and applications of protein nanoparticles.# Prepared protein (hen lysozyme) nanoparticles and tested for its cytotoxicity on drosophila cell lines# Prepared nanoparticles were subjected as nanocarrier for plasmid DNA, eGFP in drosophila cell linesTechnical Skills Developed:Protein aggregation at alkaline pH, Fluorescence Microscopy (Inverted and TIRF), Mass Spectroscopy (MALDI-TOF), Atomic Force Microscopy, Cell Culture Techniques, Biochemical assays (MTT assay), Protein labelling with fluorescent probes like fluorescein, dansyl, Cy3 and Cy5
Visiting Researcher
This is a Summer Research Fellowship awarded by Indian Academy of Sciences (IAS).Studied the role of Accessory Proteins and Ionic Liquid Substrate Interactions towards Improving Enzymatic Efficiency of Biomass Degrading Enzymes.Technical Skills Developed:Protein purification, Enzyme kinetics and Enzyme activity assays
Research Intern
# Involved in collection and biochemical and histopathological processing of biopsies from leprosy and tuberculosis patients# Studied the role and expression of chemokine genes (CXCL8 and CXCL10) in development and progression of leprosyTechnical Skills Developed:General molecular biology techniques (RNA extraction, RNA quantification, Electrophoresis and RT-PCR) and Tissue processing for histopathology
