TOPIC:- Identification and Molecular characterization of Myxobacterial species Isolated from thar desert.DESCRIPTION:- I have completed my project work in 5 months of period. The gram negative fruiting growth pattern myxobacteria was isolated using SOIL DUNG BATING method and further screened by streak plate and method, One omore method was followed that was E.coli. bating. The better result was observed in first method. After isolation of myxobacteria confirmation was carried out by biochemical tests and microscopic observations. Pure culture of the myxobacteria was used for the extraction of the chromoomal DNA and amplification of the DNA with myxo specific primer to confirm the presence of myxobacteria on 1.2% agarose gel. The PCR products was further sent for 16S rRNA gene sequencing. After sequencing I got various numbers of unculturable and culturable unknown species and some species was already sequenced and known.Scope : Myxobacteria is the species which has the antimicrobial activity against different diseases. Its one of the species stigmatella having the antimicrobial activity which produces anticancerous drug which can be used for the treatmen tof the cancer.

I worked on PCR based marker system like SSR, ISSR, RAPD Molecular marker. In this I have selected the Pearl millet seeds having very good growth in hot condition and kept for growth for 1 week. After growth I have islated the DNA and follwed the PCR process for diffrent marker systems. I have sed different primers for diffrent specie which were universal primers. After getting DNA band by following diffrent marker system I have idenetified the band size of the pearl millet and then analysed for the phylogeny.
