This project is investigating the structure of cell walls and looking at a novel protein that allows the cell wall to rearrange in previously unseen ways. Research AssistantMoredun Research Institute May 2022 - Mar 2023Penicuik, Scotland, United KingdomAs the research assistant in a molecular virology lab studying Ovine Pulmonary Adenocarcinoma (OPA), my role is to provide lab-based support to the clinical trial of OPA infection. The overall aim of the study is to develop a model of pulmonary lung cancer that can be used in human diagnostics, alongside the development of a vaccine for the Jaagsiekte respiratory virus (JSRV) that causes OPA in sheep and goats. I was responsible for planing and executing my own work to deliver results that carry the project forward. I analysed, interpreted and discussed the scientific data my work produces with my PI on a regular basis and suggested next steps.Techniques and and skills used in this role include: manual RNA extraction from exhale breath condensate, lung fluid and nasal swabs; RT-qPCR using ABI7500 and QuantStudio5 thermocyclers; Post mortem removal of lungs and lymph nodes from sheep and preparing tissue for histology and DNA extraction; Purification of plasmids from E.coli; DNA extraction from lung tissue; Handling, manipulating, updating and maintaining large data sets; Working at ISO9001 GLP standards. -
Research AssistantMoredun Research Institute May 2022 - Mar 2023Penicuik, Scotland, United KingdomWorking as Research Assistant for the Jonhe's group, I provide technical support in the diagnosis of Johne's disease in ovine and bovine herds in Scotland.My day to day work involves culturing M. avium subspecies paratuberculosis (MAP) on plates, slopes and in suspension, maintaining these cultures and producing growth curves of each species to determining its growth cycle; developing and optimising a phage plaque assay as a diagnostic tool for identifying MAP infection and quantifying MAP culture viability; liaising with farmers to set up surveillance projects on sheep flocks to detect MAP and parasite co-infection in live farms; On-farm work sampling animals and soil for active infections; Working at ISO9001 GLP standards and applying QA requirements to diagnostic testing.I was heavily involved in a gut loop infection study, managing the cultures used to inoculate the gut loops, determining their optimal infection growth stage; producing the inoculum; assessing its viability on the day of infection; and preparing the gut tissue, post mortem, for analysis, such as histology, bacterial recovery and RNA sequencing. I was the deputy Point of Contact for the CL2 lab in which several groups are working on a variety of projects and was jointly responsible for maintenance of the lab and its equipment, and ensuring it operates within the Code of Conduct and H&S guidelines. This also included reviewing and updating the SOPs and Risk Assessments for all MAP work. -
Molecular TechnologistNhs Lothian Dec 2020 - Apr 2022Edinburgh, Scotland, United KingdomThe purpose of the brand new virology lab was to test for positive SARS-CoV-2 samples in populations of Care Home staff, prison staff, hospital staff, clinical trials and pre-op or ward admitted patients by RT-qPCR during the Covid-19 pandemic.I was an integral participant in the setup and running of the lab, being one of the first 6 people to open the lab use to my CL3 Covid-19 experience. I was heavily involved in calibrating new equipment, testing and validating new procedures, performing analyser comparisons for UKAS accreditation, and training new staff. I was actively involved in management communications regarding creating and improving SOPs. My role involved supervising and problem solving in different areas of the lab, requiring in-depth knowledge of each part of the process to troubleshoot errors on the large scale automation liquid handling Hamilton robots, extraction robots and thermocyclers, and keep the lab functioning at a high throughput capacity while maintaining the highest standards of quality control procedures. I had to keep up to date with any changes that occurred while I was working elsewhere in the process.As a key member of the training team, I was responsible for training new staff members. As each new group of healthcare assistants started in the lab, I was responsible for their education and training in safely handling and deactivating live virus samples in Microbiological Safety Cabinets (MSC).I was responsible for assessing competencies of other lab members and updating the competency records to accurately reflect abilities of each individual. -
Covid Support ScientistMoredun Research Institute Nov 2020 - Dec 2020Penicuik, Scotland, United KingdomAs part of an NHS subcontracted lab, I worked under tight deadlines during the Covid‑19 pandemic performing virology diagnostics with a 24 hour turn around time. This was done in a Containment Level 3 (CL3) laboratory using Class II Microbiology Safety Cabinets (MBSCs)an strict adherence to Standard Operating Procedures (SOPs), and included full training in CL3 pathogen handling and waste disposal. All work was carried out at ISO 9001:2015 standards of quality assurance and under Good Laboratory Practice.The process involved working with live virus samples and highly toxic chemicals, and required an understanding of the COSHH and safety procedures associated with these chemicals as well as what to do in the case of an accidental chemical or biological spill.A high level of team work was required to coordinate each stage of the operation. Each job linked directly to the next stage of the process and required a high level of good communication to ensure it proceeded smoothly. I often took over work from other team members to allow for breaks or handed work over, meaning that all work had to be done methodically to be easily followed by someone else. At all times, it was important that I was aware of what was going on in the lab around me and how my work fitted in to the overall process. -
