High-Throughput Screening/Nuclear ReceptorsConstructed a series of peptide fusion vectors that allowed for the co-expression of native or phage derived co-activator (TIF-2, SRC, PGC-1) or co-repressor (NCOR) like motifs with an NR linker (nuclear receptor) of choice. The expressed protein contained a HisTag, the peptide of choice, glycine and NR of choice. Three encoded thrombin sites allowed for removal of the His tag and glycine linker during purification thus providing protein suitable for crystallography. Co-expression in this manner resulted in increased expression levels and aided in NR crystallography pursuits.In Assay Development I performed the GCNF Gal4 Reporter ultra high-throughput screening assay. The assay, performed in a 384-well format used multiple robots to screen a 10,000 compound library over 9 nuclear receptors for possible activities. The screening assays were analyzed using binding curves. The curves were scrutinized and catalogued into a complex computer data base called A-base. Results of these screens were then used to improve structures for future compounds for further development in various scientific groups.
