At UDOPP I am working as a project team member in antbacterial projects within ENABLE (European Gramnegative Antibacterial Engine). My responsibilities in the projects includes to perform differnt ADME in vitro assays, and report and present these data at project meetings.The in vitro assays are: - plasma protein binding.- metabolic stability in hepatocytes and liver microsomes.- metabolic stability in microsomes with phase 2 co-factor UDPGA and pore-forming peptide alamethicin. UDP-glucuronosyl transferase, UGT, dependent metabolism.- kinetic and thermodynamic solubility.- CYP induction with probe substrates for CYP1A2, CYP2B6 and CYP3A4.- CYP inhibition with probe substrates for CYP1A2, 2C8, 2C9, 2C19, 2D6 and 3A4.- CYP3A4 time dependent inhibition.

Since 2010 I have been a core-team member in several preclinical projects within neurology, virology and oncology. I was responsible for the in vitro DMPK strategy of the projects, including the coordination of the in vitro work, external in vitro studies at CRO, delivery of data, presentations within the department and project groups and writing reports.My focus the last years have been on strategy work concerning "Nucleotide prodrug approach in Oncology" and on permeability and metabolism of nucleotide and nucleoside prodrugs in intestine, liver (hepatocytes) and peripheral blood mononuclear cells.During 2000-2010 I was involved in building up the ADME activities at Medivir, especially in vitro assays measuring intestinal permeability (Caco-2 assay), possible impact of P-gp on the ability of small molecules to pass the blood-brain barrier (MDCKII-MDR1) and stability ( clearance intrinsic) of small molecules in liver (primary hepatocytes).In projects where brain exposure was critical, transport experiments with bovine brain endothelial cells were used.During these years I also set-up the liver perfusion technique to generate primary hepatocytes from mouse and rat, and assays to measure degree of inhibition of small molecules on different uptake transporters in the liver.I was also supervisor for several students and scientific support for less experienced colleagues.
