The same enzyme in Anaerobic Methanotrophs is responsible for oxidation of Methane to carbon dioxide. The forward reaction is extremely thermodynamically favored. How does an enzyme reverse a thermodynamically favorable reaction to activate the highly stable C-H bond of methane is an intriguing and challenging research question. I study the anaerobic oxidation of methane by MCR in vitro, isolated from a methanogenic archaea using HPLC, EPR, ENDOR, MCD and XAS spectroscopy. -

Postdoctoral Research FellowUniversity Of Michigan, Ann Arbor Jan 2004 - Sep 2006930 N University Dr. Ann Arbor Mi• Performed cloning, over expression (22L fermentation), purification( FPLC) and characterization (UV-vis enzyme activity assay) of the mutant adenosylcobalamin (AdoCbl) dependent enzyme glutamate mutase (Glm-ES). Designed HPLC enzyme assays to monitor steady state kinetics of the movement of tritium from the enzyme to substrate to product and back. Formulated a mechanism for substrate binding, activity and the relative energy barriers between the various species in the enzyme pocket.• The non-enzymatic reaction kinetics of adenosyl radical formation was studied with ESI-MS and GC-MS to determine the presence of hydrogen tunneling by following the formation of the quenched adenosyl radical, 5’-deoxyadenosine.• 2-thioglutarate a competitive inhibitor of the enzyme (Glm-ES) was designed and extensive EPR studies was done to detect the resultant radical species in solution. Simulations (collaborators) confirmed the presence of a stable thioglycolyl radical species. The above studies facilitate the understanding of enzyme active sites, which in turn can be used to engineer these sites towards different substrates and vice versa. The knowledge gained can be utilized to design drugs based on specific enzyme targets. -

Residents of Sakinaka, Mumbai slums were monitored for their exposure to heavy metals like lead and air pollutants like nitric oxide, sulfur dioxide, trioxide etc. A detailed study was conducted and the data presented to the Principal investigator. Unwilling participants had to be convinced of the importance of this data collection and the effect it could have on their quality of life. Developed good interpersonal relations with the subjects which facilitated in effective data collection.
