About Anjali Patwardhan, Ph.D.

Bio-analytical chemist by training with specific interest in physical methods to tackle research problems. Open to working in varied areas of research i.e. fundamental research, applied research, translational research. Interested in teaching and mentoring young scientists. Ability to adapt to different environments and work on multiple problems at the same time. Independently worked on nucleic acid biochemistry during PhD and followed it with research in protein biochemistry during postdoctoral studies. Extremely driven and excited to work on newer goals. Special expertise in applying previous knowledge to tackle new challenges to arrive at better solutions. Worked effectively as part of a team and/or led and motivated team members to complete the task at hand. Effectively organized and prioritized tasks to deliver individual research goals in a timely manner. Successfully completed the Training and Education Standard issued by the NIH for conducting clinical research within the intramural research program Excellent communication skills, written and spoken.Skills and Expertise: • Next Generation Sequencing library preparation for DNA-Seq and RNA-Seq followed by analysis of Sequencing data. • Real time quantitative PCR.• Cloning, Transformations, Protein expression (fermentation on 22L scale) and purification (FPLC and PAGE), Enzyme kinetics, RP-HPLC, NMR, EPR, Radioactive labeling, RNA/DNA syntheses, RT-PCR, PCR, UV-visible spectroscopy, Flourimetry, Rapid chemical/freeze quench EPR, Calorimetry.• Developing methodology/assays for separation of reactions products from complex biological (DNA/RNA and protein enzyme) reactions.• Assisting in the writing of grant proposals and manuscripts for peer reviewed publications.• Routine maintenance, function and training of personnel on above analytical instruments.

Methyl Coenzyme M reductase is the final enzyme responsible for the release of all biological methane as part of the Methanogenesis metabolic pathway of methanogenic archaea. The same enzyme in Anaerobic Methanotrophs is responsible for oxidation of Methane to carbon dioxide. The forward reaction is extremely thermodynamically favored. How does an enzyme reverse a thermodynamically favorable reaction to activate the highly stable C-H bond of methane is an intriguing and challenging research question. I study the anaerobic oxidation of methane by MCR in vitro, isolated from a methanogenic archaea using HPLC, EPR, ENDOR, MCD and XAS spectroscopy.

Succcessfully initiated the Next Generation Sequencing (NGS) project in the lab and prepared genomic libraries for DNA and mRNA libraries for RNA Seq from three different strains of ecoli to determine the changes in the sequence of genomic DNA in the absence of uracil excision pathways, namely knockouts of the genes ung (Uracil DNA glycosylase) and dut (dUTPase).

    • Performed cloning, over expression (22L fermentation), purification( FPLC) and characterization (UV-vis enzyme activity assay) of the mutant adenosylcobalamin (AdoCbl) dependent enzyme glutamate mutase (Glm-ES). Designed HPLC enzyme assays to monitor steady state kinetics of the movement of tritium from the enzyme to substrate to product and back. Formulated a mechanism for substrate binding, activity and the relative energy barriers between the various species in the enzyme pocket.• The non-enzymatic reaction kinetics of adenosyl radical formation was studied with ESI-MS and GC-MS to determine the presence of hydrogen tunneling by following the formation of the quenched adenosyl radical, 5’-deoxyadenosine.• 2-thioglutarate a competitive inhibitor of the enzyme (Glm-ES) was designed and extensive EPR studies was done to detect the resultant radical species in solution. Simulations (collaborators) confirmed the presence of a stable thioglycolyl radical species. The above studies facilitate the understanding of enzyme active sites, which in turn can be used to engineer these sites towards different substrates and vice versa. The knowledge gained can be utilized to design drugs based on specific enzyme targets.
  • Center For Environmental Sciences And Engineering, Iit Powai
    Senior Research Scientist
Center For Environmental Sciences And Engineering, Iit Powai Jun 1996 - Mar 1997
    Powai, Mumbai, India
Procured and analyzed samples of suspended air particles/pollutants for their heavy metal content along with mentoring a junior assistant for the purpose. Residents of Sakinaka, Mumbai slums were monitored for their exposure to heavy metals like lead and air pollutants like nitric oxide, sulfur dioxide, trioxide etc. A detailed study was conducted and the data presented to the Principal investigator. Unwilling participants had to be convinced of the importance of this data collection and the effect it could have on their quality of life. Developed good interpersonal relations with the subjects which facilitated in effective data collection.
