"Accurate determination of the amount of exosomes secreted by human reticulocytes by integrating colloidal nanoplasmonics and light scattering techniques" In University of Brescia I learnt how to isolate extracellular vesicles using colloidal system and to characterize them with different assays such as western blot, Atomic Force Microscopy (AFM) and colorimetric nanoplasmonic assay (CONAN).The aim of this project is to develop a robust and accurate method for the determination of the basic colloidal properties, including molar concentration, size distribution and stability in common buffers, of exosomes isolated from culture media of human reticulocyte cell lines and from THP1 cells and porcine serum. I compared and inter-calibrated two techniques, Nanoparticle Tracking Analysis (NTA) and a colorimetric nanoplasmonic assay (CONAN), in order to obtain a robust quantification of exosome formulations. In particular I focused on reticulocytes-derived exosomes (rex) because they have an important role in Malaria pathogenesis. Two main themes of my thesis were the inter-calibration and the isolation and characterization of exosome derived from different sources (reticulocytes, THP1 cells and porcine serum). I used different exosome isolation protocols: sucrose gradient and size exclusion chromatography. Exosomes were characterized by bead-based assay (using an alternative FACS analysis) for exosomal markers and with BCA assay for the presence of exogenous proteins (contaminants). I inter-calibrated CONAN and NTA, which have different physical-chemical principles, and the fact that was possible is an important result. The inter-calibration curve allows to choose the method that better fits exosome preparation. I provide the first steps in quantification of rex.
