Department Of Plant Pathology And The Genome Center

- Worked with a team of 4 to 8 members as bacterial pathology lead for pathogen virulence and plant innate immunity projects. - Coordinated collaborations with other research groups in multiple locations to analyze bacterial virulence factors and plant interaction. - Introduced new pathogenecity systems for analysis of rice hydathode infection, tomato-Xanthomonas euvesicatoria interaction, and subtle virulence differences. - Developed new proteomic approaches to detect sulfated, processed, and small secreted bacterial peptides.- Implemented E.coli system with an expanded genetic code to express sulfated proteins- Designed and planned new projects for NIH, NSF, USDA grant proposals.- Mentored 3 graduate, 2 undergraduate, and 3 high school students. My research successfully identified and characterized a small peptide molecule from Xanthomonas oryzae pv. oryzae, which binds and activates rice immune receptor XA21. As a results, I published 7 peer-reviewed papers and obtained one U.S. patent.

Molecular Biology And Microbiology

- Characterized ADP-ribosyltransferases (ADP-RT) from tomato pathogen Pseudomonas syringae that are translocated through a Type III secretion system (T3SS) into plants cells and function as virulence factors that suppress plant innate immunity. Performed in vitro ADP-RT assay, Protein-RNAs binding assay (gel shift assay), and pathogenesis assay.- Characterized GRP7, a glycine-rich RNA-binding protein, that are modified by an ADP-RT, HopU1 from P. syringae and contributes plant innate immune response. Generated Arabidopsis transgenics expressing GRP7 and HopU1, and assessed their immune response and immune-related hormone production against P. syringae infection.- Performed soybean transformation to over-express Arabidopsis GRP7 or soybean GRP7 homologs and tested their enhanced immunity with a challenge of P. syringae. - Demonstrated that for the first time the T3SS-mediated suppression of innate immunity of rhizosphere Pseudomonas, which produces an antibiotic to suppress wheat disease pathogens in agricultural soils.- Contributed to work showing bacterial T3S is inhibited by plant immune response Induced by the recognition of conserved bacterial molecule.

Demonstrated the role of Trehalose-6-phossphate synthase (TPS) in plant abiotic stress resistance including drought, cold, salt, and heat stresses. - Performed a microarray analysis to identify genes differentially expressed in the Arabidopsis over-expressing TPS.
