Analysis revealed that efficient mobility of LAT clusters, during T cell activation, is dependent on a change in protein composition as clusters move through different actin architectures. • Developed colocalization software package, ColocP2C, designed to measure the colocalization between two imaged molecules when one displays a punctate appearance and the other displays a more continuum appearance. Applied this tool to multi-channel imaging of the receptor, CD36, and its downstream effector, Fyn. Analysis revealed that CD36 nanoclusters in F-actin-rich regions were colocalized with Fyn at all times, regardless of the presence of its ligand, TSP-1. • Developed a transient mobility analysis tool, termed “Divide-and-Conquer Moment Scaling Spectrum” (DC-MSS), designed to detect changes in molecule movement from particle tracking data. Applied this tool to single-particle tracks of the transmembrane protein CD44 on the surface of macrophages, revealing actin cortex-dependent transient mobility changes. Show less
