PhD thesis entitled “Characterization of ADP ribosylation factor-like GTPase 15 (ARL15) in rheumatoid arthritis synovial fibroblast cell line using an integrated omics approach”- Unraveled ADP ribosylation factor-like GTPase 15 (ARL15) function in rheumatoid arthritis (RA) - Employed ARL15 knockdown (siRNA) and overexpression (plasmid-based) approaches in RA synovial fibroblast (MH7A) cells.- Employed an integrative multi-omics approach to decipher the ARL15's role in RA (gene expression microarray, transcriptome, Real-Time PCR, pulldown, and proteomic analyses).- Identified 25 potential interactors of ARL15 in RA synovial fibroblasts using integrative pathway analysis.Other associated work during Ph.D. apart from thesis work- Synovial fluid & synovial tissue collection – Coordinated with doctors for RA and Osteoarthritis (OA) patients’ synovial fluid and tissue samples collection at AIIMS, New Delhi (2015 - 2017).- Synovial Fluid & Tissue Processing (RA & OA) for culture of synovial fibroblast cells (2015 – 2017).- FACS - FACS analysis of cell surface marker (CD90/Thy1) to identify primary synovial fibroblast cells and apoptosis (Annexin V/PI) assay (2015 – 2017).- Animal work - CIA mice model to compare ARL15 monoclonal antibody vs. TNF inhibitor on disease remission & joint pathology (2017 – 2018).- Schizophrenia case-control study – Microarray (Agilent Sureprint G3) analysis of 44 samples for copy number variations and SNPs (2016).
