1) The Mouse Ear: A new model system to study hair cycle regulation:My main project during my time at the Plikus lab. I carried out: - performing procedures on mice under anaesthesia- tissue collection and imaging (Gross imaging and Fluorescent microscopy)- EdU pulse-chase experiments- EdU assay development and optimization2) In-vivo hair characterization studies using mouse models- breeding and handling in vivo mouse models- sample collection and characterization- sample imaging and measurement- data collection and analysis- presentation of analyzed data- collaboration with the Mathematics department for mathematical modeling and statistical analysis 3) Optimization of dermal and epidermal cell collection from Mouse back skinThis mini-project was carried out using a special LEF-RFP/K14-H2B-GFP mouse model> Mouse tissue collection (back skin)> Tissue processing, to obtain single cell suspension> FACS to separate out the dermal and epidermal components of skin4) Trained undergraduate students in the essential techniques required for the above projects

During my time at Allergan, I was mainly involved in the following three projects:1) Gene Expression Studies Testing Novel Ophthalmic Compound - Cell Culture of primary human RPE cells and stable transformed RPE cell line (RPE-19) - RNA extraction and cDNA preparation from cells treated with compound - qPCR to measure gene expression of cAMP responsive genes - qPCR data analysis and interpretation2) In vivo Experiments Using Wild-Type and Knockout Mice Testing Compounds For Photoreceptor Protection - Mouse model using bright light to induce photoreceptor damage - Measured damage induced by light both visually by Heildelberg Retinal Angiograph (HRA) for auto-fluorescence and Electro-Retinal Angiogram (ERG) for functional response. - Whole eye enucleation for histological analysis, posterior eye-cup preparation and retina dissection for other studies.3) Performed experiments Testing Novel Dermatology Drug - In vivo drug testing using Golden Syrian Hamsters for effect on the sebaceous gland - Prepared drug prior to treatment - Quantitative measurement of dermal thickness response after drug treatmentPresented the results and progress of these projects to the Vice-Presidents of Allergan and other team members at the monthly meeting
