About Anushka Gupta

I am a Scientist in the Assay Development, Molecular Biology team at 10x Genomics where I am currently working on building new methods for tissue profiling with transcriptomics and protein co-detection at single-cell level resolution. Previously, I graduated from the UC Berkeley-UCSF Graduate Program with a PhD in Bioengineering where I focused on developing novel methods for characterizing the human adipose tissue at the single-cell level. My specific fields of interest include microfluidics engineering, single-cell analyses, bioinformatics, and optical microscopy. I have expertise in spatial multi-omics, single-cell and single-nuclei RNA-seq, short-read sequencing, design and development of molecular assays, live-cell microscopy, and fluidics engineering. I am also proficient in establishing bioinformatic pipelines for the analysis of high-dimensional scRNA-seq data.I am passionate about technology and methods development to enable rapid discovery of fundamental biology implicated in health and disease and am looking forward to working in this area in my next opportunity!

    • Developed a microfluidic workflow, μCB-seq, for paired imaging and RNA-sequencing of single-cells. Optimized sequencing chemistry for improved gene detection sensitivity while reducing library preparation costs by 50x. Automated the entire operational workflow using laboratory robotics.• Implemented computational and statistical methods to reveal transcript enrichment and detection biases in single-nuclei RNA-seq as compared to single-cell RNA-seq. Developed a novel normalization framework to account for said detection biases while analyzing sequencing datasets.• Designed and performed multiplexed, time-course, snRNA-seq experiments (~ 100,000 nuclei) to study human adipogenesis. Developed a sample preparation workflow for efficient isolation of intact nuclei from fragile specimens. Established an end-to-end bioinformatic pipeline for the analysis of acquired high-dimensional snRNA-seq datasets.
