Project: Bacteriophage to control the Pierce's Disease (PD) caused by Xylella fastidiosa in grapes. Program Involved:- Conducting field trials in California (Santa Clara, Sonoma and Solano County) and Texas (Uvalde County) on evaluation of a phage-based biocontrol system for Pierce’s Disease (PD) of grapevines for California EPA, OMRI and DPR approval. - The Xylella and bacteriophage treated plant samples (weed, sharpshooter, and petiole) were collected from the fields (California & Texas) and analyzed for the movement and quantification of Xylella and phage cocktail using PCR and qPCR.- Genomic sequencing and characterization of phage and bacteria- Determined uptake and transmission of Xylella fastidiosa by vector glassy-winged sharpshooters- Production of Master Phage Bank (MPB) and Master Cell Bank (MCB)- Construction of Xanthomonas pilA deletion mutants to determine primary phage receptor sites on the bacterial cell surface- Developed, executed, and optimized upstream (fermentation, scale-up, characterization) and/or downstream (centrifugation, and tangential flow filtration) in accordance with GLP requirements

Project Title I: “Development of primers for improved detection of plant pathogens & pests”Programs Involved: - Development of molecular diagnostics tools in the Texas Plant Disease Diagnostic Lab TPDDL). - Detection of pathogens in plant samples by traditional and molecular methods (PCR and qPCR). - Development of primers for improved detection of pathogens and pests. - Conduct pathogenicity tests for different pathogens on different crop species. - Screening the efficacy of the biopesticides for managing bacterial diseases in greenhouse. - Identification of biotic and abiotic disorders of horticulture, fruits, and vegetables crops.Project Title II: “Development of bacteriophage plaque assay for Liberibacter”Synopsis: The main objective of this project is the development of a phage-based biocontrol system for citrus greening (HLB) and zebra chip (ZC) diseases. We are screening to identify the bacteriophages and tailocins that can kill the Liberibacter crescens which is close to Las pathogen in citrus and Lso pathogen in potatoes. Project Title III: “Development of new technologies for improved field detection and identification of the ZC pathogen”Synopsis: Development of loop-mediated isothermal amplification procedure (LAMP) for an improved, simplified PCR-based detection system to detect the ZC pathogen, ‘Candidatus Liberibacter solanacearum’ (Lso). LAMP technology promises reliable, rapid, and cost effective method for detecting Lso pathogens in psyllids and field-grown potato. Project Title IV: “Whole genome sequencing, assembly and annotation of Pseudomonas syringae pv. syringae” Synopsis: I have succeeded in completing full annotated genomes using the Next Generation Sequencing (NGS) platforms (454 pyrosequencing and Illumina Solexa). This experience lays the foundation for me to comparative genomic analyses to related Pseudomonas spp. pathogens, including unique regions, mechanisms of host range, virulence factors, and toxin synthesis.

Project I - Title: “Endophytic bacteria for biological system management of Radopholus similis the key nematode pest of black Pepper (Piper nigrum L)”. Synopsis: During my Ph.D. research studies in Mangalore University in India, I characterized the endophytic bacterial flora from black pepper (Piper nigrum L.) and screened these bacteria for antagonism against Phytophthora capsici and Radopholus similis, two major pathogens causing foot rot and slow decline of black pepper plants. The biocontrol agents produced more than 70% disease suppression in black pepper and this shows the potential for successful disease control in the field. Subsequently, I also developed a rapid and reliable single tube duplex PCR for simultaneous detection of P. capsici and R. similis infecting black pepper. Project II - Title: “Application of Microorganisms in Agriculture and Applied Sectors (AMAAS) – Nutrient Management, PGPR and biocontrol”. - Isolation and identification of biocontrol and Plant Growth Promoting Rhizobacteria (PGPR) from black pepper and ginger.- Screening for identification of efficient and effective biocontrol bacterial strains for suppressing pathogens (Phytophthora capsici, and Pythium myriotylum) and pests (Meloidogyne incognita and Radopholus similis).- Biochemical and molecular characterization of biocontrol and PGPR isolates.- Evaluation of effects of the biocontrol and PGPR isolates under nursery and field condition. - A strategy for production of disease free plantlets of black pepper in nurseries.- Scaling up and formulation of promising isolates of PGPR.
