Loss of tissue architecture is one of the main hallmarks of cancer (tumor grade). and tumour grade has been correlated with patient outcome. Moreover PTEN loss has been associated with high grade cancers in colorectal, prostrate and breast cancers. In colorectal cancer, we have previously shown that loss of PTEN lead to decreased CDC42/aPKC/PAR6 polairt complex pathway. My project is to find out a link between PTEN and CDC42 and to understand how PTEN regulates CDC42 activity.

I was looking at resistance mechanisms against MEK inhibitors in KRAS and BRAF mutant patients in colorectal cancer. We found out that JAK/STAT pathway is one of the main resistance mechanisms. So we combined MEKi with JAKi or STAT3 antisense, measured the efficiency of this combination therapy and compared it to conventional drugs in BRAF and KRAS mutant cell lines.
