They consist among others in the excision of precisely bounded sequences, the IES (Internal Eliminated Sequences). This excision is carried out by the domesticated transposase. Piggymac (Pgm) and the repair of DNA double strand breaks introduced by Pgm is assured by the Non Homologous End Joining pathway (NHEJ). The Ku heterodimer is a necessary partner of Pgm for the introduction of double strand breaks. My graduate internship was aiming to characterize the Ku/Pgm interaction. To do so, I developed in the laboratory the two-hybrid technic in the yeast Saccharomyces cerevisiae, which will allow the screening of loss of interaction mutants between Ku and Pgm.
