As a Facilities Director of both the Macromolecular Crystallography Facility (MCF)/ Crystallography Automation Facility (CAF) and the Physical Biochemistry Instrumentation Facility (PBIF), I am an experienced research scientist in both chemical biology and biochemistry, with expertise in structural biology and X-ray crystallography techniques for soluble and membrane proteins. The MCF/CAF was founded to facilitate and encourage research endeavors in protein X-ray crystallography and structural biology. Managing this facility has allowed me to gain and strengthen skills in implementing advanced high-throughput automation robotics in crystallization experiments. In fact, one of my main roles as facilities director of the MCF/CAF is to maintain the integrity of robotic function and reliability. I have performed screening utilizing the ARI Phoenix DT/RE and performed optimization with the Rigaku Alchemist DT. With the ARI Crystal Gryphon LCP, I helped researchers implement experiments toward crystallization of membrane protein utilizing lipidic cubic phase. Automated imaging/inspection occurred with the Rigaku Minstrel HT and I aided researchers in analysis with the CrystalTrak application. Subsequently, I have experience utilizing a home source Rigaku RU-H2R R-Axis IV X-Ray generator as well as remote access to Beamline 4.2.2 at the Advanced Light Source (ALS) for screening and data collection My knowledge of the instruments and research allow me to aid and train users in order to provide a centralized resource for training and education in latest crystallographic and structural biology techniques. I developed the graduate course, Biophysical Analysis of Macromolecules, offered to junior graduate (1st/2nd year) students. The objective of the class is to foster an understanding of the theory, application, and limitations of available instrumentation housed in the PBIF. These instruments are commonly used to solve questions inherent to many biochemical/biophysical research endeavors. These are invaluable but underutilized approaches to simply and rigorously characterize macromolecular interactions, multimeric status of complexes, order of assembly of macromolecules and rates of important processes. Students taking the course benefited in having direct hands-on exposure to each instrument covered through practical experiments. In addition, having such experience will aid in familiarity to the PBIF (as well as the MCF) and knowledge of experimental capabilities inherent to instruments covered within the course. Experiences in the class through practical exposure may hopefully be extrapolated to usage and implementation of instruments by students in their research endeavors toward the completion of thesis projects.

The work as a research associate involves analysis and characterization of human folate receptors (hFR) by taking the information gained from the solved/refined structural models and applying these insights to mutant studies of the receptor. Key interactive residues in ligand binding have been identified through my doctoral work to achieve atomic resolution structures of hFRs in states that mimic folate uptake and biological trafficking for both folates and antifolates. The mutant constructs were designed and expressed transiently and in mammalian Chinese hamster ovary (CHO) cell lines. These mutant hFR proteins were characterized through biophysical means such as ITC to evaluate binding affinity compared to wildtype protein. These mutants will also be structurally characterized in comparison to our available FR structural models in hopes of improving rational drug design based on structural information rather than strictly on empirical principles for increased efficacy in chemotherapeutics.

My doctoral work solved the atomic resolution structures of human FRs (hFR) in states that mimic folate uptake and trafficking for both folates and antifolates. Membrane bound folate receptors (FRs) are cell surface glycosyl-phosphatidylinositol (GPI)-anchored glycopeptides. Two membrane bound FRs have been identified and characterized to date, FRα and FRβ. FRα is a tumor marker overexpressed in several cancerous malignancies of epithelial origin. Consequently, FRα is widely recognized as a potential target for the selective delivery of a broad range of therapeutic agents to tumors. FRβ is upregulated in activated macrophages involved in inflammatory disease. Physiological traits have made FRs a relevant drug vector due to high affinity binding and the potential to transport a chemically diverse range of folate conjugates, antifolate drugs, and immunological agents. Design of drugs for FR-mediated therapy has been entirely empirical and efficacy is still limited due to transport via the ubiquitously expressed reduced folate carrier (RFC), causing dose limiting toxicity to normal tissue. The structural models of my doctoral work are imperative to improving rational drug design based on structural information rather than strictly on empirical principles for increased efficacy in chemotherapeutics.
