My tasks consisted of participation in analytical method development for new products, and execution of analytical method validations and verifications, forced degradation studies and photostability studies. The project involved the development of potent and selective probes against histone decaylase 11 (HDAC 11) for investigation of this isoform as a potential drug target. The approach was to synthesize macrocyclic non-ribosomal peptides that would be selective towards HDAC 11. For this synthesis, first a non-natural amino acid was chemically modified with a moiety that was supposed to selectively target HDAC 11, and then this amino acid was incorporated into the peptide along with other natural amino acids through SPPS. Finally, the linear peptide was cyclized in solution to yield the desired macrocyclic peptide. The potency and selectivity of the final compounds were evaluated by performing enzymatic biochemical assays.Through this project I became accomplished at organic synthesis having performed a variety of chemical reactions, compound purification by flash chromatography and prep-HPLC, compound identification by LC-MS, UPLC and NMR, and pharmacological evaluation of compounds by enzymatic inhibition assays.Grade: 12/12Supervisor: Professor Christian Adam Olsen
