My thesis research focused on validating the pathogenecity of variants identified by Next Generation Sequencing-based molecular diagnosis of patients with retinitis pigmentosa (RP) /leber congenital amaurosis (LCA) and using CRISPR-targeted knock-in and knock-out mouse models to elucidate the pathogenicity of candidate disease causing genes. In addition to characterizing the phenotype of disease conditions in animal models, my thesis also focused on identifying the molecular mechanisms by which loss of a gene’s function results in blindness. The translational aspect of my work utilized gene editing and AAV-based approaches to develop a successful, clinically validated gene therapy treatment for a rare, inherited form of blindness. Further continuation of these clinical efforts from bench to bedside will help prevent blindness in children and adults.
