Developing a new methodology for amplification of a fluorescent label using a heterosandwich complex (2 different ligands which bind, ACh-PTS). An endogenous ligand, acetylcholine, was used as a structural model to develop a technique using encapsulated (fluoro media) liposomes as the amplified fluorescent response which exhibits an anchored molecule which binds to one end of the heterosandwich complex (binding to PTS). The other half of the complex binds to acetylcholine (ACh) receptors with a high affinity, allowing a cascade of binding events to occur. 1) ACh moiety binds to ACh receptor on a cell. 2) PTS moiety binds to liposome 3) liposome is encapsulated with fluorescent response, allowing an unnaturally large response compared to molecules bound to the cell.
