About Aurora (Rory) Blucher, Phd
Computational biologist with domain expertise in druggability and precision cancer therapeutics. My methods expertise grew from a foundation of mathematical modeling training to encompass biological pathway and network modeling approaches. In my research contributions, I use this systems-level view across data types to ask hard questions about disease progression, therapeutic vulnerabilities, and development of drug resistance. As a data scientist and connector, my role is to quantify and communicate the evidence that will allow cross-functional teams to develop and accelerate therapies to achieve durable patient responses.Previously I was a postdoc in Gordon Mills' Lab at the Knight Cancer Institute, where I worked on understanding druggability and prioritizing combinations for triple-negative breast cancers and ovarian cancers. This provided me with experience in a highly translational and team-science setting, where wet and dry lab members developed strategy for analysis and validation in close collaborations. During my time at OHSU, I was an active computational contributor to precision oncology research groups across campus, including the Head and Neck Precision Oncology team, the Beat AML team, and the SMMART precision clinical trials team. Here at Recursion, you’ll find me on the Drug Discovery Inference and Oncology teams, where I work to identify our strongest leads in cancer druggability questions and push these discoveries into compelling and promising clinical drug programs.

Given the uncertainty inherent in drug-target interaction information, I developed the Cancer Targetome evidence framework for tiering supporting evidence of drug-target interactions. This allowed for prioritization of drug-target interactions in accordance to strength of evidence and enabled quick triage of patient 'omics alterations with respect to candidate therapeutic vulnerabilities. Furthermore, using a probabilistic graphical modeling approach, I integrated functional mutation calls for AML patient biopsies with quantitative drug-target information from the Cancer Targetome into a pathway-based modeling framework. This systems approach provided a view of patient tumors that highlighted pathways expected to be jointly highly impacted in tumors and most likely to be druggable.
