Thesis Title: Development of Novel Fluorescent Nanoparticles based Diagnostic Assay for theEarly, Rapid and Accurate Detection of HIV Infection.Our course of research involved pioneering studies on nanoparticle engineering and development of effective sensing strategies to address the sensing needs of the resource limited settings we have ultimately developed a fluorescent nanoparticles based immunosensor for the ultrasensitive detection of analytes with affordable sensors.Fluorescent SiO2, ZnO nanoparticles and Carbon quantum dots were synthesized by chemical methods and studied for their structural and morphological properties by TGA, XRD, IR, UV, SEM and DLS. These fluorescent nanomaterials were used to develop nanoparticles based immunosensors for the ultrasensitive detection of HIV detection. The project involved in the identification of various promising targets for the identification of HIV co infected with TB. In the first step bio-conjugation of Europium encapsulated polystyrene nano particles with streptavidin was done by by cross linking with various amino groups, avidin-biotin, disulphide coupling and to use them in the fluorescent based immunoassay. These bioconjugated nanoparticles used to detect both antigen and antibodies of HIV and other co infections like TB in our study. We could develop a microfludic chip for the simultaneous detection of HIV and TB which can be developed into a point of care kit.
