Supervisor: Susana Chuva De Sousa LopesActivities/achievements:- Research focus: the derivation of germ cells from pluripotent stem cells.- Discovered an essential role for the extracellular matrix in primordial germ cell (PGC) specification, allowing high efficiency PGC generation in vitro.- Genome editing of hiPSC using CRISPR/CAS9 to generate endogenous reporter lines - Analysis of scRNAseq data to map the signaling environment in fetal gonads- Development of improved 3D culture methods for reconstituted fetal gonad- Writing of scientific papers and presentation at international conferences.- Techniques used: Stem cell culture, Stem cell differentiation to (primordial) germ cells, CRISPR/Cas9, RNA-seq (bulk and single-cell), FACS, molecular cloning and microscopy.

- Research Focus: Elucidating disease mechanisms of congenital protein trafficking disorders (primarily microvillus inclusion disease and Wilson disease) in stem cell models and polarized cell lines.- Independent set up of the complete workflows for hiPSC-based research, which was not present in the department at the time. This included: generation of iPSCs line from patient-derived somatic cells, subsequent maintenance and culture of hiPSC, and the differentiation of hiPSC to hepatocytes.- Generation of bile canaliculi forming hepatocytes from Wilson Disease patients to evaluate potential treatments.- Knock-out of MYO5B and STX3 in hiPSC, CACO-2 and HepG2 cells using CRISPR/CAS9- Molecular cloning and introduction of various patient mutations in HepG2 using lentiviral transduction. This work formed the basis of discovering a new mechanism for the hepatic symptoms of microvillus inclusion disease.- Writing of scientific papers (with 3 published as first author), and presentation at conferences.- Supervision of technician and students.- Techniques: Stem cell culture, Stem cell differentiation to hepatocytes, lentiviral transduction, CRISPR-CAS9, confocal microscopy, Western blot, qPCR, molecular cloning.
