In June 2023, I joined the pediatric Leukemic Research Group as a Research Assistant specializing in Genomics. I'm excited to embrace new challenges and contribute to the team's efforts. At TTCRC, I'm delving into diverse areas of high throughput Genomics. My tasks within the Genomics workflow encompass a wide range, including SNP array analysis, mRNA sequencing, and the bio-banking of bone-marrow and peripheral blood samples from important leukemic patients. I'm also responsible for tasks such as DNA and RNA isolation, endpoint PCR, multiplex PCR, fragment analysis, and Sanger sequencing. I've gained proficiency in using advanced instruments like the 3500 Genetic Analyzer, NextSeq 550, proflex, step one plus, Nanodrop, and Qubit. My primary focus revolves around establishing an efficient workflow to enhance risk assessment for patients. This involves identifying TP53 mutations through Sanger sequencing, detecting IKZF1 deletion via PCR and fragment analysis, and pinpointing ERG deletion in IKZF1del patients. Additionally, I work on identifying various high-risk CNVs using SNP array analysis. These efforts align with our overarching goal of improving diagnostic and treatment approaches for better patient outcomes.

We aim to look into the significance of this microbial protease HAP in autophagy induction in this study. It has been previously proved that HAP activates PAR-1 by cleaving the N-terminal sequence of "PFISEDASGY and produce a 6 amino acid long peptide (PFISED) that activates PAR-1 and subsequently induces PAR-1 mediated apoptosis. In this present study, we found that HAP can trigger autophagy in cancerous cells through PAR-1 mediated pathway and our observation also suggests that ‘PFISED’ peptide alone is capable of inducing autophagy and causing cell death in cancer cells.
