I am currently seeking Lab Technician positions in Orlando Florida. During my stay at Dr. Chakrabarti's lab, I wrote proposals and got funding for two undergraduate research projects. I participated in a paid program for undergraduate research called Research and Mentoring Program for two years. I presented in the Showcase of Undergraduate Research and won judge's award about my pancreatic cancer research project. I gained hands on experience in lab in the following:Western-blotting, PCR, QPCR, Tissue Culture, troubleshooting, Cloning, transfection, transformation, Migration assay, making gels, and protein extractions. My projects while in lab:As an undergraduate, I was working on three projects throughout my junior and senior year. I worked on and completed a cloning project of creating an inducible expression system for a long non-coding RNA in PLVX-tet-one vector to be used in animal studies. With the supervision of Dr. Thomas Andl and help with CRISPR primer design, I made a knockout system for a long non-coding RNA. This consisted of initial CIRSPR RNA primer design, annealing, vector digestion, purification, ligation, followed by testing the best combinations of CRISPR primers using Alt-R Genome editing kit. Alt-R Genome editing kit allowed us to observe whether the transient transfections using our knockout vectors caused a mutation. After verification, we performed a double transfection and cell sorting to cause a double deletion which causes a knockout. The results thus far showed that the knockout severely inhibited the grow of the cells. The cells grew too slowly and were too fragile to survive freezing. Hence we are currently performing another round of sorting with the inducible vector as a rescue mechanism. My final project was about characterizing FGD4, a guanine exchange factor, in relation to pancreatic cancer progression. I preformed over-expression studies in a pancreatic cancer cell line to see its effects on cell migration, EMT markers, and a cytoskeleton transduction pathway.
