Lab of Mitchell Kronenberg, President and Scientific DirectorProjects:• Regulation of glycolipid-driven Th-polarization by CD1d. Investigated the paradox of iNKT activation with small molecule glycolipid agonists. Discovered that the key to Th1/Th2-polarization is in the stability of the glycolipid’s contact with the MHC class I-like antigen presenting molecule, CD1d, not in the manner in which the iNKT cells are activated. Manuscript in preparation to be sent to a leading scientific journal.• Diverse ligands can activate iNKT cells but an O-linked galactose is optimal. Determined hierarchy of affinity and off-rates for CD1d:TCR complexes, correlated hierarchy with ERK1/2 phosphorylation and in vivo activation.Managerial experience:• Supervised and trained five individuals -- one visiting medical student, three undergraduates, and one technician; including mentoring one undergraduate honors thesis.

Arts, Associate Professor of MedicineCase Western Reserve University, Cleveland, OH 1997-1998Project:• Analysis of patients from Uganda: Molecular Epidmiology of HIV-1 and possible effects of Mycobacterium Tuberculosis on Viral Quasispecies. Initiated project to examine whether HIV-1 heterogeneity is altered in HIV-1-infected individuals with TB. Blood samples were obtained from patients with active TB (HIV/TB patients) and HIV-1-infected patients (HIV patients) in Kampala, Uganda. The C2-C3 region of the HIV-1 envelope gene (env) was amplified from lysed peripheral blood mononuclear cells (PBMCs), and then subject to sequencing, clonal-quasispecies analysis and heteroduplex tracking analysis (HTA). Concluded that an increase in HIV-1 heterogeneity may be associated with a TB-mediated increase in HIV-1 replication.
