About Ben Hock

-I presently lead the cross-functional translational sciences/research early development teams for an AAV5 gene-therapy and an enzyme-substitution therapy. These teams give me the opportunity to contribute as a scientist and interface with corporate governance and other partner teams as a team leader.-Functional responsibilities within Clinical Immunology include immunogenicity strategy (mitigation approaches, varieties of assays, nonclinical approach), interpretation and reporting of results, and response to regulatory questions on immunogenicity.-My present interests are (1) analysis and presentation of immunogenicity impact (on safety, efficacy, and exposure), (2) support strategies for ERTs, peptide therapeutics, and gene-therapies (3) clinical and nonclinical assays to unpack varieties of hypersensitivity (ex. IgE, tryptase, complement), and (4) immunotoxicity assessment tools (ex. T-dependent antibody response studies).-During my tenure at Amgen (2010-2017), I was responsible for validated immunoassays [clinical and non-clinical (GLP)]. Analytes included anti-drug antibodies, total IgE, tryptase and anti-KLH antibodies (immunotoxicology endpoint). Platforms included MSD, Biacore, and ImmunoCAP. I also represented the clinical immunology group on mAb, BiTE, peptide, and oncolytic virus program teams.-Since joining BioMarin in 2017, I've been able to support the interpretation and reporting of additional exciting modalities- Enzyme replacement (and substitution) therapies, AAV gene-therapies, and oligonucleotide therapeutics.-Skilled in a wide range of molecular techniques including: DNA-microarray, analysis of protein-DNA interactions by chromatin immunoprecipitation (ChIP) in-vitro and in-vivo, design and analysis of quantitative PCR assays for genomic and mRNA targets, viral mediated gene-transfer, siRNA targeting of gene expression, transfection-reporter assays, protein overexpression and purification, and protein interaction analysis by surface plasmon resonance (Biacore).-Experience in diverse fields including immunology and physiology.-Publications: https://pubmed.ncbi.nlm.nih.gov/?term=Hock%20MB&sort=dateKewords: Immunogenicity, anti-drug antibody (ADA), immunoassay, immune complex, hypersensitivity, complement, IgE, MSD, Biacore.
