Knowledge Driven Approaches and Machine Learning Improve the Identification of Clinically Relevant Somatic Mutations in Cancer Genomics Advisors: Elaine R. Mardis PhD, Obi L. Griffith PhDMy thesis work centered on developing methods an resources to make better use of the vast amounts of cancer genomic data. I have created a web resource (http://docm.genome.wustl.edu) that outlines curated cancer mutations that were found to be biologically or clinically important in the scientific literature. I have leveraged this resource to create a knowledge-based variant detection methodology that ensures important variation was not missed by traditional sequencing analysis workflows (published in Nature Methods). Additionally, I have used machine learning to automate the manual review of called sequence variants (Published in Nature Genetics). I have also contributed extensively to 12 additional publications involving biological data science, crowdsourcing, cancer genomics, bioinformatics and human genetics.
