As a PhD candidate focusing on electron ptychography, I am exploring its application in biological sciences, moving beyond its traditional use in material sciences. This involves not only pushing the boundaries of the technique but also innovating in its application to new areas of research.In my work, I integrate a mix of commercial and custom-built components to assemble efficient electron ptychography systems. This process involves bringing together high-speed direct electron detectors from providers such as Dectris and the Paul Scherrer Institute (PSI), utilizing comprehensive software such as SerialEM, and incorporating cutting-edge algorithms like py4DSTEM package. These components are carefully combined to create systems that fulfill the demanding requirements of biological 4D-STEM research.Beyond using commercially available products, I also develop tailored solutions to meet specific research needs. For instance, the custom scan generator I designed for our Talos and Titan Krios microscopes is a testament to this ability. The quality of the ptychographic images captured with this equipment aligns with, or surpasses, current standards for biological specimens in the ptychography field.My primary aim is to advance imaging techniques and data analysis in biological electron microscopy, with a special emphasis on phase-sensitive imaging. By doing so, I hope to contribute significantly to the growth and evolution of this important scientific field of computational imaging.
