About William Mallet

As a scientist and a drug discovery leader, nothing gives me greater pride than learning that a patient and her doctor entrusted her care to a drug that I helped develop. I am driven to discover truly innovative therapies that will give patients greater hope and longer lives. Benefiting from great colleagues and committed organizations, I have gained deep expertise in all aspects of drug discovery, from concept to clinic. My goal is to put my learnings to practice in an emerging company, building a motivated Research team to bring novel drugs to people in need.Key roles: Research lead for discovery and development of biologic therapeutics for cancer and fibrosis. Project head and lead scientist on multiple successful antibody-drug conjugate (ADC) programs for oncology. Team builder and leader across cultures, functional areas, and company stages. Thought leader in oncology biologics, with special emphasis on antibody conjugates and bi-specific antibodies.

William Mallet Work Experience Details
  • Pedalion Bio Llc
    Founder And Managing Partner
Pedalion Bio Llc Jul 2024 - Present
I established Pedalion Bio to provide consulting support for pre-clinical drug discovery and development. Our focus is on biologics for cancer, leveraging my broad experience in the field. We target early-stage and emerging companies, through internal pipeline build and partnering discussions.
  • Bolt Biotherapeutics, Inc.
    Senior Director, Target Validation
Bolt Biotherapeutics, Inc. Mar 2022 - Jul 2024
    Redwood City, California, Us
Bolt Biotherapeutics is develops myeloid-activating biologics, including immune-stimulating antibody conjugates (ISACs) to trigger a broadly active immune response to cancer. At Bolt, I was responsible for selecting molecular targets and leading programs from inception into clinical development. I led two external partnerships and our internal CEA project; helped establish and validate the ISAC platform; supervised our Antibody Engineering team; and contributed to Bolt strategy as a member of leadership teams.
  • Bolt Biotherapeutics, Inc.
    Staff Scientist, Target Validation
Bolt Biotherapeutics, Inc. Nov 2018 - Mar 2022
    Redwood City, California, Us
  • Maverick Therapeutics
    Director, Pre-Clinical Pharmacology
    Maverick Therapeutics Jun 2017 - May 2018
    Brisbane, California, Us
My role focused on bringing novel conditionally activated bi-specific T cell engagers (COBRAs) to the stage of IND-enabling studies. I was responsible for lead molecule selection and characterization (in vitro and in vivo); design, coordination, and interpretation of in vivo efficacy and PK studies; IHC method development and antigen profiling; and diverse CRO relationships.
  • Gilead Sciences
    Senior Research Scientist 2
    Gilead Sciences Jun 2014 - Apr 2017
    Foster City, Ca, Us
My focus was on pre-clinical development of biologics for fibrosis, oncology, and inflammation. My responsibilities included target discovery and validation, plus determining the best applications for novel biologics formats. I led a team to identify a candidate CD3-redirected T cell engaging bi-specific, and I developed multiple project proposals for novel and established therapeutics.
  • Novartis
    Senior Investigator I
    Novartis May 2010 - May 2014
    Basel, Baselstadt, Ch
I led a project team seeking to discover and develop novel antibody-drug conjugates (ADCs) as cancer therapies. Having worked in this field for twelve years, I have seen first-hand the huge potential that ADCs have to make a difference for patients. From my base in Emeryville, I collaborated with Novartis associates around the world, pulling together experts in diverse areas of biologics, chemistry, pharmacology, DMPK, toxicology, and translational medicine to bring important new therapeutic options to patients with greatest need.
  • Genentech, Inc.
    Scientist
    Genentech, Inc. Feb 2002 - Apr 2010
    South San Francisco, California, Us
I led a project team to identify new tumor antigens for antibody-based therapeutics, particularly the development of antibody-drug conjugates as cancer drugs. The research in my lab spanned the pre-clinical spectrum, from target discovery through in vivo efficacy, using recombinant DNA, protein biochemical, and cell biological methods. Additionally, I collaborated with Genentech's Antibody Engineering, Bioinformatics, Pathology, Translational Oncology, Safety Assessment, Pharmacokinetics, Clinical Science, and Commercial groups. During my eight years at Genentech, my lab made key contributions to our understanding of the parameters governing antibody-drug conjugate activity and safety and our assessment of tumor antigen targets.
  • Genesoft, Inc.
    Staff Scientist
    Genesoft, Inc. Dec 1999 - Nov 2001
I worked with a team of molecular biologists, biochemist, and medicinal chemists to evaluate GeneSoft's novel DNA-binding small molecules for regulating gene expression. I developed a microscopy screen to quantify the nuclear accumulation of these molecules in live cells, providing data that our chemists applied to improve the delivery of the molecules to the target DNA.
  • Weill Cornell Medical College
    Postdoctoral Fellow
Weill Cornell Medical College Jul 1996 - Dec 1999
    New York, Ny, Us
I worked in the lab of Dr. Frederick Maxfield in the Department of Biochemistry. My studies focused on the quantitative analysis of post-endocytic protein sorting pathways, using fluorescence microscopy and biochemistry. In the course of this work, I characterized two novel membrane trafficking pathways.
