About Binu Antony

I’m a researcher interested in developing methods through synthetic biology and chemical ecology approaches for sustainable insect pest management. I hold a Ph.D. in Biological Sciences, with over 15 years of research experience in entomology, chemical ecology, functional genomics, and synthetic biology. My research experience in RNAi, functional genomics (multi-omics, heterologous expression, etc.), high-throughput sequencing and analyses, transgenics, functional characterization of genes, electrophysiology, and cell culture. From 2012 until Sept 2023, I worked at King Saud University-Riyadh and led a research group on chemical ecology and functional genomics. My research specialized in functional characterizing insect pheromone biosynthesis genes and engineering to produce pheromones in microbial systems. We demonstrated the pivotal role of the pheromone gland-specific fatty acyl-CoA reductases (pgFAR) and an E12 desaturase in making species-specific signals in moths, genetically engineering yeast and incorporating these biobricks to produce cost-effective pheromone bioproduction. Our research targeting chemoreceptor proteins involved in odorant detection insects was demonstrated using an RNAi knockdown of OR-Orco, and it proposed that disrupting chemical communication would be a promising approach for controlling insect pests. Combining OMICS-driven approaches, including RNAseq, loss-of-function through RNAi, and Drosophila expression, we have characterized the first RPW pheromone receptor and a kairomone receptor. We concluded a research project on developing OR-OBP-based biosensors for the early detection of palm weevil infestation in the date palm field. We run two major grants on developing olfactory protein-based biosensors, genome engineering, and olfactory communication disruption. We achieved a key milestone in unraveling the molecular basis of olfaction R. ferrugineus. We identified the chemosensory gene family proteins involved in odorant detection from R. ferrugineus (Antony et al., 2016). We demonstrated using an RNAi technique to knock down olfactory gene expression (Soffan et al., 2016). We proposed that disrupting the palm weevil olfactory system had been a promising approach for controlling this pest. We identified the Odorant binding protein (OBP) specific to the aggregation pheromone ferrugineol of RPW (RferOBP1768). We demonstrated that the RferOBP1768 gene can be silenced with RNAi, leading to pheromone anosmia, thereby providing a solid baseline for further development of palm weevil management programs (Antony et al., 2018). By a combination of OMICS-driven approaches, including RNAseq, loss-of-function through RNAi, and transgenic (Drosophila) expression methods, we have characterized the very first RPW pheromone receptor (RferOR1) (Antony et al., 2021). The discovery of the pheromone receptor won the prestigious Khalifa International Award for Date Palm and Agricultural Innovation 2022 - Distinguished Innovative Studies and Modern Technology. We characterized an odorant receptor tuned to several ecologically relevant palm-emitted odors.

Our task was to develop and deliver updated information on arthropod pest management for tea crops in the Northeastern Indian states of Assam and West Bengal (the largest tea-growing region globally). I worked closely with several tea garden extension scientists and cooperative extension teams in Assam's four major tea-growing areas (Cachar, North Bank, South Bank, and Upper Assam). Our team is dedicated to highlighting an important, newly emerged major pest of tea, commonly known as tea looper caterpillars: Biston spp. and Hyposidra spp. In early 2000, Looper caterpillars emerged as severe pests of tea, and at least four species were found to invade the entire tea-growing belt of northeast India. Sometimes, mixed broods appear in various tea gardens in Assam. We proposed several new initiatives to tackle the looper problem and supportive evidence that the situation of a looper outbreak resulted from climate change in northeast India (Antony et al., 2012). Through field extension activities, we reported a very appealing note that the beginning of this century was marked by the increased report of looper invasion, and in the last two years, looper attacks have been reported from almost all Assam and North Bengal tea gardens. We reported novel nucleopolyhedrosis virus (NPV) infecting loopers (Antony et al., 2011), proposed new biopesticides, and prompted the tea industry and practitioners to adopt the microbial management of loopers.
