About Blake Chaffee

Worked in an NIH funded lab using the ocular lens as a model to reveal molecular mechanisms regulating developmental processes influencing cell induction, proliferation, survival, and epithelial cell to fiber cell differentiation. Initiated multiple projects involved with DNA methyltransferase activity in the developing ocular lens, and formulated the ideas funding an R21 NIH grant. Led projects focusing on the antagonism between the tumor suppressor, Phosphatate and Tensin homolog (Pten), and Fibroblast growth factor (Fgfr) signaling in the lens, ocular glands, corneal epithelium, and peri-ocular epidermis. Characterized the first post-mitotic role of cyclin dependent kinase 1 (Cdk1) in the process of denucleating lens fiber cells. Currently elucidating the mechanism by which Fgfr signaling controls DNaseIIβ transcription in differentiating lens fiber cells.

    Germline mutations in PTEN represent one of the primary causes of Cowden’s Syndrome (CS), PTEN Hamartoma Syndrome, Bannayan-Riley-Ruvalcaba Syndrome, and Proteus syndrome. Somatic mutations in PTEN have been discovered in nearly every cancer. Additionally, germline mutations in PTEN are found in about 10% of children with Autism spectrum disorder (ASD) and macrocephaly. My work focuses on the neurological development resulting from PTEN mutations that lead to ASD and macrocephaly.
